Increased Akt pathway signaling has been shown to be directly correlated with increased rates of glucose metabolism observed in cancer cells versus normal cells. These observations have led to the proposal that inhibition of Akt signaling would inhibit glycolysis and increase hydroperoxide production which would preferentially kill tumor cells versus normal cells via oxidative stress. The current study shows that inhibition of the Akt pathway inhibits glucose consumption and induces parameters indicative of oxidative stress such as glutathione disulfi de (%GSSG) and thioredoxin reductase (TR) activity in human head and neck cancer (HNSCC) cells. A theoretical model to explain the results is presented and implications for the use of Akt pathway inhibitors in combination with glycolytic inhibitors and/or manipulations that increase prooxidant production are discussed.

Glutamine relieves oxidative stress through PI3K/Akt signaling pathway in DSS-induced ulcerative colitis mice

Objective(s): Ulcerative colitis (UC) is a kind of complex immune disease, and a major cause of destruction of intestinal barrier and oxidative stress in this field. In this paper, glutamine (Gln) was believed to offer protection against oxidative stress injury in colitis mice.Materials and Methods: Thirty mice were randomly assigned int...

Salvianolic acid B improves insulin secretion from interleukin 1β-treated rat pancreatic islets: The role of PI3K-Akt signaling

Background and Objective: Oxidative stress induced by proinflammatory cytokines such as IL-1β plays a major role in β-cell destruction in diabetes type 1. Salvianolic acid B (Sal B) is a polyphenolic compound with antioxidant and protective effects. Thus, objective of this study was to assess the protection exerted by Sal B on isolated rat islets exposed to IL-1β and to investigate an underlyin...

Role of oxidative stress in pathogenesis of metabolic syndrome

The metabolic syndrome (MS) recognized as a major cause of type 2 diabetes and cardiovascular diseases, has become one of the major public health challenges worldwide. The pathogenesis of the metabolic syndrome is multiple and still poorly understood. No single factor has yet been identified as an underlying causal factor. There is a growing belief, however, that obesity, specially visceral obe...

Role of Oxidative Stress in Modulating Unfolded Protein Response Activity in Chronic Myeloid Leukemia Cell Line

Background: Recently, it has been revealed that tyrosine kinase inhibitors (TKIs) act through inducing both oxidative and endoplasmic reticulum (ER) stress in chronic myeloid leukemia cells. However, ER stress signaling triggers both apoptotic and survival processes within cells. Nevertheless, mechanisms by which TKIs avoid the pro-survival effects are not clear. The aim of this study was to ev...

Effect of maternal variable stress on oxidative status and glucose metabolism in pubertal male rats

Introduction: Metabolic disorders are affected by negative stress experiences in the early stages of life. Accordingly, in this study, the effects of stress during pregnancy on oxidative status and glucose homeostasis in pubertal male Wistar rats were investigated. Materials and Methods: After pregnancy, female rats (200±30 g) were divided into 2 groups (6 rats per group) of stress and non-stre...
